Chemotherapy of viral infections

  1. Docosanol
    • Inhibit adsorption and fusion
    • Modify cell membrane by making them rigid
  2. Adamantes
    • Inhibit viral uncoating
    • Mimic H+
  3. Acyclovir, Ganciclovir, Cidofovir
    • Inhibiting viral nucleic acid replication through sugar modification
    • Mimics dGTP DNA chain terminator
  4. Brivudin, Idoxuridine, Trifluridine
    • Inhibiting viral nucleci acid replication through base modification
    • Causes mis-translation --> competitive inhibitor of DNA polymerase
  5. Foscarnet
    • Inhibit viral nucleic acid replication through non-nucleoside DNA polymerase inhibitor
    • Prevents cleavage of pyrophosphate from dNTPs during chain elongation process
  6. Ribavirin
    • Inhibit viral nucleic acid replication through RNA polymerase inhibition
    • monophosphate inhibits inosine monophosphate dehydrogenase --> depletion of GTP
    • di & tri phosphate inhibits viral RNA polymerase
  7. Raltegravir
    Inhibition of integrase activity
  8. Zanamivir, Oseltamivir
Inhibit viral penetration and budding through neuraminidase/sialidase activity
  9. Interferon
Inhibiting viral processing through IFN-alpha and IFN-beta
  10. Antisense agents
Synthetic oligonucleotide expected to seek target site --> hypridize with it --> inhibit the normal function ofthe particular DNA or RNA segment.
