Quiz
what is the name of the microscope
Bright field light microscope
what does the bight field light microscope do
produces dark image against light background
who is the bright field light microscope most comminly used by
biology students
what is the bright field light microscope used for
to view preserved stains
which means dead specimen with dye on it
what is the framework of the microscope
arm
base
head which rotates
what is the stage
fixed platform that supports the microscope slide
what is the mechanical stage
allows horizontial and vertical movement
allows to center the specimen
what is the light source
its build into base
round knob allows you to control intensity of light
what is the focusing knob
concentrically arranged knobs on side of microscope that brings specimen into focus
what are the focusing knobs on the microscope
coarse adjustment
fine adjustment
what is the coarse adjustment
larger
outer knob
rough image
what is the fine adjustment
smaller
inner knob
fine image
what are the 3 types of lenses
ocular lens
objective lens
condenser
ocular lens
eye piece
binocular because it has 2
magnification of 10x
objective lens
the 4 lens that are attached to rotating nosepiece that allows them to move over a slide
what are the 4 objective lens
4x
10x
40x
100x
4x
scanning
shortest
red
10x
low power
yellow
40x
high dry
blue
100x
oil immersion
white
need to use drop of oil
lenses need to touch the oil for it to work
condenser
below stage
lens that collect and direct the light from the lamb to the slide being studied
what does the condenser do that the ocular and obective lens doesn't
does not effect the magnifying power
what is the part within the condenser and what does it do
diaphragm which controls amount of light that reaches the slide
also known as iris diaphragm
what are the 2 jobs of the microscope
magnification
resolution
what do we mean when we say one of the microscopes job is magnification
ability to enlarge objects
what do we mean when we say one of the microscopes job is resolution
capacity to distinguish or separate two adjacent objects as separate
what are the two phases that magnification occurs in
the objestive lens magnifies and then the ocular magnifies again to form an image
what is the formula to calculate total magnification
power of objective X power of ocular= total magnification
what is the purpose of oil
to keep the light into image
prevents refraction of light
what is the total magnification of each objective lens
4x=40x
10x=100x
40x=400x
100x=1000x
what are the steps to use the microscope
center specimen
start with low power objective (10x)
turn coarse adjustment knob until image appears
bring object into focus with fine adustment
once image is visible progess to next objective
what do you never want to do after focusing the image with fine adjustment
do not readjust coarse objective knob
what sould you do before moving objective to oil immersion
place a drop of oil on the slide
go half way between objectives, put the drop if oil, then finish sliding objective into place
should be touching oil
transport
always carry with 2 hands
one hand on base
other hand on arm
do not let cord dangle to aviod tripping
what sould you clean lenses with
lens paper and solution if needed before and after use
clean oil immersion last
cleaning the microscope
remove the slide
clean lenses
lower stage
rotate to scanning objective because less damange
before putting microscope away have teacher check it
wrap cord
replace dust cover
put under proper location
