Cell Bio Test #2

  1. Describe IgSF (immunoglobulin superfamily)
    • *cell-to-cell adhesion receptor class
    • *many perform homophilic interactions
    • *Members:CAMs-cell adhesion molecules
    • *proteins named by their well-organized loops
    • *Why Important? -involved in bundling of neurons in the embryonic nervous system
  2. What are Cadherins?
    • Important group of adhesive glycoproteins found in the plasma membrane
    • **Distinction!: these require calcium to function. (IgSFs do not) - Ca2+ binds to and stabilizes the conformation and allows them to mediate cell-cell adhesion
    • characterized by similar domains or "repeats" in their extracellular domain
    • Why Important? -play important roles in embryonic development. -involved in helping neuronal cells to form bundles and establish synaptic connections
  3. Lectins
    • Carbohydrate-binding proteins
    • promote cell-cell adhesion by binding to a specific sugar or sequence of sugars exposed at the outer cell surface
    • Usually have more than one carbohydrate-binding site, therefore, they can bind to carbohydrate groups on two different cells, linking two cells together
  4. 1.Selectins and 2.Leukocyte Adhesion
    • 1.cell surface glycoproteins that mediate interactions of leukocytes with endothelial cells
    • 2. Leukocytes roll along the walls of blood vells. During inflammation, they attach to the wall of a blood vessel near the inflammation and migrate through the blood vessel to the inflammation site. The initial attachment is mediated by binding of selectins on the leukocyte to carbohydrates on the surface of the endothelial cells and vice versa. When the leukocyte stops rolling and begins to invade the blood vessel, integrins on the surface of the leukocyte and an IgSF protein called ICAM on the surfaces of endothelial cells mediate this interaction
  5. Blood type determination
determined by a specific carbohydrate side chain present on a glycolipid of the red blood cell plasma membrane.
