341 - Anti-nicotinic Drugs

• Competitve antagonist of neuronal nicotinic receptor
• Used iv
• Rapid onset of action, but it shows tachyphylaxis (progressive decrease of effectiveness with continual use)
• Malignant Hypertension: severe syndrome characterizied by a very high diastolic BP
• Postoperative Hypertension: less severe syndrome characterized by a high systolic and diastolic BP.

• Competitive antagonist
• Blocks mostly muscle nictonic receptor (leads to skeletal muscle relaxation) and has modest ganglion nicotinic blocking activity

• Non-competitive ganglion nicotinic blocker
• Open channel block of the nictonic receptor (can't be overcome by increasing ACh)
• Crosses blood-brain barrier
• Hypertension Treatment: drug causes bothersome side effects at therapeutic doses
